Cellular mechanisms whereby quiescent stem cells sense tissue injury and transition to an activated state are largely unknown. Quiescent skeletal muscle stem cells (MuSCs, also called satellite cells) have elaborate, heterogeneous projections that rapidly retract in response to muscle injury. They may therefore act as direct sensors of their niche environment. Retraction is driven by a Rac‐to‐Rho GTPase activity switch that promotes downstream MuSC activation events. These and other observations lead to several hypotheses: (1) projections are morphologically dynamic at quiescence, (...) providing a surveillance function for muscle damage; (2) quiescent projection dynamics are regulated by the relative balance of Rac and Rho activities promoted by niche‐derived cues; (3) projections, particularly their associated filopodia, sense tissue damage via changes to the biomechanical properties of the niche and/or detection of signaling cues released by damaged myofibers; and (4) the dynamic nature of projections results in a population of MuSCs with heterogeneous functional properties. Her radical and unorthodox ideas are important not only because they anticipated the more tolerant, ecumenical, and optimistic philosophy of the Enlightenment, but also because of their influence on Leibniz. Anne Conway’s Principles of the Most Ancient and Modern Philosophy, first published in 1690, is probably the most ambitious contribution to early modern metaphysics by a woman writing in the English language. This beautifully prepared edition makes Conway’s treatise available to twentieth-century readers in an accessible English translation of the 1690 Latin text—itself a translation of an original English manuscript that has long been lost.

We argue that the experience of knowing and having the truth about oneself known in the context of therapy is not an end in itself; rather, it is important because the trust engendered by this experience opens one up to learning about one’s social world and finding better ways to live in it. We consider the consequences of a lack of epistemic trust in terms of psychopathology.

Clinician-scientists play a valuable role in bridging the gap between patient care and clinical research. However, there are no clear guidelines that describe how to handle the day-to-day ethical and methodological questions that arise when navigating the competing responsibilities of practitioner and researcher—a situation that can easily leave the clinician-scientist confused and frustrated. This article outlines common ethical and methodological concerns for the clinician-scientist when developing, implementing, and analyzing research in practice, and offers practical suggestions to address these issues. How do ethics show up in the everyday behaviors and conversations of researchers in a scientific laboratory? How does the microcosmic culture of the laboratory shape researchers’ understandings of scientific ethics? We, an interdisciplinary team representing anthropology, computer science, and rhetorical studies, investigated these questions in two university STEM labs. Similar to previous work mapping out the epistemic cultures, we sought to understand the ethical cultures of these research groups. We observed their lab meetings for several months and conducted interviews (...) with members of the research laboratories. Ultimately, we recognized two distinct ethical cultures within the two laboratories: one focused on the relationship between science and social justice, and the other focused on clean data and accuracy as an ethical issue. These two distinct ethical cultures were discernable through ethnography, close reading of laboratory members’ language, as well as broad-scale topic modeling. Our project complements previous studies of ethics in science that have focused on codes of ethics or student curricula as ways in which scientists develop an understanding of scientific ethics. Our project demonstrates that everyday interactions between the members of scientific research labs additionally shape, at a decidedly local level, how scientists understand and ultimately approach ethical decision-making in the scientific research process. Infant and young child electrophysiology studies have provided information regarding the maturation of face-encoding neural processes. A limitation of previous research is that very few studies have examined face-encoding processes in children 12–48 months of age, a developmental period characterized by rapid changes in the ability to encode facial information. The present study sought to fill this gap in the literature via a longitudinal study examining the maturation of a primary node in the face-encoding network—the left and right fusiform gyrus. (...) Whole-brain magnetoencephalography data were obtained from 25 infants with typical development at 4–12 months, and with follow-up MEG exams every ∼12 months until 3–4 years old. Children were presented with color images of Face stimuli and visual noise images that served as Non-Face stimuli. Using distributed source modeling, left and right face-sensitive FFG evoked waveforms were obtained from each child at each visit, with face-sensitive activity identified via examining the difference between the Non-Face and Face FFG timecourses. Before 24 months of age the face-sensitive FFG M290 response was the dominant response, observed in the left and right FFG ∼250–450 ms post-stimulus. By 3–4 years old, the left and right face-sensitive FFG response occurred at a latency consistent with a face-sensitive M170 response ∼100–250 ms post-stimulus. Face-sensitive left and right FFG peak latencies decreased as a function of age, and with an adult-like FFG latency observed at 3–4 years old. Study findings thus showed face-sensitive FFG maturational changes across the first 4 years of life. Whereas a face-sensitive M290 response was observed under 2 years of age, by 3–4 years old, an adult-like face-sensitive M170 response was observed bilaterally. Future studies evaluating the maturation of face-sensitive FFG activity in infants at risk for neurodevelopmental disorders are of interest, with the present findings suggesting age-specific face-sensitive neural markers of a priori interest. Cloth, $93.75.In this detailed examination of Henry More’s psychological explanation of enthusiasm, Daniel C. Fouke persuasively argues that previous discussions of seventeenth-century attitudes toward (...) enthusiasm have been marred by misplaced attempts to categorize them in terms of “rationalism,” “empiricism,” and the “new science.” As he says, “a central problem in every account is that anti-enthusiasts and their targets simply refuse to ally themselves according to commitments which, to our eye, should sort them into well-defined genealogies, neat schools, and clear enmities. Clear battle-lines fail to emerge, and any portrait of the typical ‘enthusiast’ never gets so far as a bare outline” (122). The ample evidence he offers to support this point appears in the three main sections of his book, in which he analyzes More’s attack on two types of scientific “enthusiasm,” alchemy, represented by Thomas Vaughan, and the mechanical philosophy, and on the religious and political enthusiasm of radical sects, represented primarily by the Quakers.Fouke points out how similar More’s own views are in many respects to the enthusiasts he excoriates, and he illustrates how tendentious and often unfair his criticisms are. For example, More viciously attacks Thomas Vaughan for his vitalistic, organic view of the natural world when he himself holds similar views. And he later attacks the mechanical philosophers for rejecting the kind of vitalism he had earlier attacked in Vaughan, while chiding them for ignoring the sympathetic effects he had detected with such repugnance in Vaughan.The same inconsistencies and misrepresentations hold true of More’s attack on the Quakers. Pediatric residents experience ethical dilemmas and moral distress during training. Few studies have identified meaningful methods in reducing moral distress in pediatric trainees. The authors aimed to determine how residents perceive ethics case discussions, whether such a program affects trainee ethics knowledge and perceived moral distress, and if residents’ perceived moral distress changes before, during, and after a discussion series. Participants included pediatric residents in a single residency program. Five separate 1-hour sessions were presented over a 5-month period. Each session (...) consisted of a case presentation by a resident developed under the guidance of an ethicist. Multidisciplinary services and content experts were present during sessions. Baseline, postsession, and final surveys were distributed to resident attendees. Open-ended responses were recorded. When comparing baseline and final responses, the only significance was increased preparedness to navigate ethical decisions (p = 0.004). A 10.2% decline was observed in perceived moral distress. An increase in ethics knowledge was observed. Residents favored case-based, multidisciplinary discussions. Residents desire more sessions, time for small-group discussions, and legal insight. Satisfaction was high with 90.7% of respondents feeling better prepared to address ethical concerns. Pediatric trainees desire case-based ethics training that incorporates small-group discussions and insight from multidisciplinary topic experts. (shrink)

It was first voiced by several of Kant's contemporaries such as Mendelssohn, Herder, Hamann, Pistorius and Eberhard who attacked the alleged subjectivism of the Critique of Pure Reason. 1 This viewpoint found its sharpest contemporary expression in the (...) notorious Garve-Feder review to which Kant responded at length in the Prolegomena. In subsequent times it has been championed by Schopenhauer, and most nineteenth century German commentators on the relation between the two philosophers. 2 In addition, it has been, and continues to be, the prevailing view of the vast majority of British writers on Kant, including, with significant qualifications, Norman Kemp Smith. 3 This tradition continues despite the fact that Kant specifically, and in no uncertain terms, repudiated this identification in the Prolegomena and the second edition of the Critique. Background: Managing HIV treatment is a complex multi-dimensional task because of a combination of factors such as stigma and discrimination of some populations who frequently get infected with HIV. In addition, patient-provider encounters have become increasingly multicultural, making effective communication and provision of ethically sound care a challenge. Purpose: This article explores ethical issues that health service providers in the United States and Botswana encountered in their interaction with patients in HIV care. Research design: A descriptive qualitative design was used (...) to collect data from health service providers and patients using focused group discussions. This article is based on responses from health service providers only. Participants and context: This article is based on 11 focused group discussions with a total sample of 71 service providers in seven US sites and one Botswana site. Ethical considerations: Ethical review boards at all the study sites reviewed the study protocol and approved it. Ethical review boards of the study’s coordinating centers, Rutgers University and the University of California at San Francisco, also approved it. The study participants provided a written informed consent to participate. Findings: HIV service providers encountered ethical challenges in all the four Beauchamp and Childress’ biomedical ethics of respect for patients’ autonomy, beneficence, justice, and nonmaleficence. Discussion: The finding that HIV service providers encounter ethical challenges in their interaction with patients is supported by prior studies. The ethical challenges are particularly prominent in multicultural care and resource-constrained care environments. Conclusion: Provision of HIV care is fraught with ethical challenges that tend to pose different issues depending on a given care environment. It is important that strong partnerships are developed among key stakeholders in HIV care. In addition, health service providers need to be provided with resources so they can provide quality and ethically sound care. (shrink)

People with lived experience are individuals who have first-hand experience of the medical condition being considered. The value of including the viewpoints of people with lived experience in health policy, health care, and health care and systems research has been recognized at many levels, including by funding agencies. However, there is little guidance or established best practices on how to include non-academic reviewers in the grant review process. Here we describe our approach to the inclusion of people with lived experience (...) in every stage of the grant review process. After a budget was created for a specific call, a steering committee was created. This group included researchers, people with lived experience, and health systems administrators. This group developed and issued the call. After receiving proposals, stage one was scientific review by researchers. Grants were ranked by this score and a short list then reviewed by people with lived experience as stage two. Finally, for stage three, the Steering Committee convened and achieved consensus based on information drawn from stages one and two. Our approach to engage people with lived experience in the grant review process was positively reviewed by everyone involved, as it allowed for patient perspectives to be truly integrated. However, it does lengthen the review process. The proposed model offers further practical insight into including people with lived experience in the review process. (shrink)

Context. The identification of the sources that reionized the Universe and their specific contribution to this process are key missing pieces of our knowledge of the early Universe. Faint star-forming galaxies may be the main contributors to the ionizing photon budget during the epoch of reionization, but their escaping photons cannot be detected directly due to inter-galactic medium opacity. Hence, it is essential to characterize the properties of faint galaxies with significant Lyman continuum photon leakage up to z 4 to (...) define indirect indicators allowing analogues to be found at the highest redshift. Aims. Long gamma-ray bursts explode typically in star-forming regions of faint, star-forming galaxies. Through LGRB afterglow spectroscopy it is possible to detect directly LyC photons. Our aim is to use LGRBs as tools to study LyC leakage from faint, star-forming galaxies at high redshift. Methods. Here we present the observations of LyC emission in the afterglow spectra of GRB 191004B at z = 3:5055, together with those of the other two previously known LyC-emitting LGRB, to determine their LyC escape fraction and compare their properties. Results. From the afterglow spectrum of GRB 191004B we determine a neutral hydrogen column density at the LGRB redshift of og =17:2 0:15, and negligible extinction. The only metal absorption lines detected are C iv and Si iv. In contrast to GRB 050908 and GRB 060607A, the host galaxy of GRB 191004B displays significant Ly emission. From its Ly emission and the non-detection of Balmer emission lines we constrain its star-formation rate to 1 SFR 4:7 M yr. 1. We fit the Ly emission with a shell model and find parameters values consistent with the observed ones. The absolute LyC escape fractions we find for GRB 191004B, GRB 050908 and GRB 060607A are of 0:35+0:10.0:11, 0:08+0:05.0:04 and :20+0:05.0:05, respectively. We compare the LyC escape fraction of LGRBs to the values of other LyC emitters found from the literature, showing that LGRB afterglows can be powerful tools to study LyC escape for faint high-redshift star-forming galaxies. Indeed we could push LyC leakage studies to much higher absolute magnitudes. The host galaxies of the three LGRB presented here have all M1600 >.19:5 mag, with the GRB 060607A host at M1600 >.16 mag. LGRB hosts may therefore be particularly suitable for exploring the ionizing escape fraction in galaxies that are too faint or distant for conventional techniques. Therefore, its meaning in biology as compared to that in physics, and the relation between the conceptions of “gradient” and “field” are studied.In physics, scalar fields, vector fields and tensor fields are distinguished. In a scalar field, the variation of the scalar in space is expressed in form of a gradient. For the whole of a scalar field with its derived gradients the term “gradient-field” may be used.In biology, especially in (...) experimental embryology, the field concept is used both in a purely descriptive sense and as a means of causal analysis. To the first group belong the kinematic fields and growth fields. The kinematic field is a vector field; the growth field is generally a tensor field, but may be treated as a gradient-field in the most simple cases. In such a field by the combined action of the scalar and the gradients a great diversity of relations may occur. Experimental evidence strongly supports the view that the subdivision of organ anlagen into smaller structural units is an autonomous process. Dalcq &Pasteels' hypothesis which says that the boundaries between the different areas into which a morphogenetic field differentiates are determined by “Threshold values” in the “potential” of the field in question, is inconsistent with our present knowledge of biochemical reaction systems. Threshold values may only be used indescribing the spatial differentiation of a morphogenetic field. It is suggested that the latter (...) concept be replaced by the concept of “optimal values” in the intensity of a morphogenetic field-specific for the various chemo-differentiations characterizing the different regions into which a morphogenetic field will split up-and that each optimal value isdeterminative for the appearance of one particular differentiation within the morphogenetic field, each differentiation being formed within a certain range of intensity of the field. On the basis of the hypothesis formulated above a “virginal” morphogenetic field will necessarily differentiate into a number of smaller fields of second order, which in their turn will split up into fields of third order, etc. ” Pattern formation” may be defined as the differentiation of a morphogenetic field into an increasing number of smaller fields of higher order of qualitative different chemical and structural composition. Since differentiation of a morphogenetic field proceeds autonomously, the capacity for pattern formation is an intrinsic property of a morphogenetic field. In the light of these considerations it is suggested that a morphogenetic field action is responsible for the first differentiation of the developing embryo into regions with predominantly ectodermal, mesodermal or endodermal differentiation tendencies. The present knowledge about the characteristic pattern formation taking place in the axial mesoderm as well as in the overlying neural plate seems to be in good agreement with the implications of the present morphogenetic field concept. It is emphasized, however, that if the present morphogenetic field concept cannot be characterized further biochemically and biophysically, it will have to be replaced by another concept which can be defined more sharply and is more easily accessible to experimental analysis. L'hypothèse deDalcq &Pasteels disant que les bornes entre les différents domaines formées par la différenciation d'un champ morphogénétique sont déterminées par des „valeurs seuil” dans le „potentiel” du champ en question, n'est pas en harmonie avec notre connaissance actuelle des systèmes biochimiques. Des “valeurs seuil” ne peuvent être employés qu'en décrivant la différenciation spatiale d'un champ morphogénétique. On prend en considération de remplacer le concept des “valeurs seuil” par le concept des “valeurs optimales” dans l'intensité d'un champ morphogénétique. Ce seraient les valeurs spécifiques aux différenciations chimiques diverses qui caractérisent les différents domaines dans lesquels un champ morphogénétique se différencie. En outre, chaque valeur optimale serait déterminative à la formation d'une différenciation spéciale dans le champ morphogénétique, chaque différentiation se formant dans un certain trajet de l'intensité du champ. Par suite de l'hypothèse formulée ci-dessus un champ morphogénétique “virginal” va nécessairement se différencier en un nombre de champs plus petits de deuxième ordre, qui à leur tour se divisent en champs d'ordre troisième, etc. ... La régionalisation peut être définie comme la différenciation d'un champ morphogénétique dans un nombre de plus en plus élevé de champs plus petits d'ordre supérieur de composition chimique et structurelle qualitativement différente. Parce que la différenciation d'un champ morphogénétique se produit de façon autonome, le pouvoir de régionalisation est une propriété intrinsique d'un champ morphogénétique. Ces considérations nous mènent à proposer que l'action d'un champ morphogénétique est responsable de la première différenciation de l'embryon en régions qui possèdent des tendences de différenciation principalement ectodermiques, mésodermiques, ou bien endodermiques. La connaissance actuelle concernant la régionalisation caractéristique qui se produit dans le mésoderme axial ainsi que dans la plaque neurale superposée paraît se conformer aux conséquences du concept de champ morphogénétique. Cependant on souligne le fait que dans le cas òu le concept actuel du champ morphogénétique ne pourra pas être plus amplement caractérisé de façon biochimique et biophysique, il faudra le remplacer par un autre concept qui puit être défini plus nettement et qui est mieux accessible à l'analyse expérimentale. Die Hypothese vonDalcq &Pasteels , dass die Grenzen zwischen den unterschiedenen Gebieten, in denen ein morphogenetisches Feld sich differenziert, durch “Schwellenwerte” im “Potential” des genannten Feldes bestimmt werden, ist unvereinbar mit unserem heutigen Wissen über biochemischen Reaktionssystemen. Der Gebrauch des Begriffs “Schwellenwert” ist nur gestattet bei der Beschreibung der räumlichen Differenzierung eines morphogenetischen Feldes. Statt dieses Begriffs wird der Begriff “Optimaler Wert” vorgeschlagen; Werte also in der Intensität eines morphogenetischen Feldes, optimal und spezifisch für die verschiedene Chemodifferenzierungen, welche die Gebiete kennzeichnen, in denen ein morphogenetisches Feld sich differenzieren wird. Jeder optimaler Wert soll dann das Erscheinen einer bestimmten Differenzierung innerhalb des morphogenetischen Feldes veranlassen, welche Differenzierung sich bildet innerhalb eines bestimmten Bereichs der Feldintensität. Auf Grund der obigen Hypothese wird ein “virginelles” morphogenetisches Feld zwangsläufig sich differenzieren in eine Anzahl kleinerer Feldern zweiten Grades; diese in Felder dritten Grades, usw. Musterbildung könnte definiert werden als die Differenzierung eines morphogenetischen Feldes in eine zunehmende Anzahl kleinerer Felder höheren Grades von qualitativ verschiedener chemischen und strukturellen Zusammensetzung, Weil diese Differenzierung autonom vor sich geht, ist das Vermögen zur Musterbildung eine innewohnende Eigenschaft eines morphogenetischen Feldes. Im Lichte dieser Betrachtungen ist es naheliegend anzunehmen dass eine morphogenetische Feldwirkung verantwortlich ist für die erste Differenzierung des sich entwickelnden Embryos in Gebiete überwiegend ektodermalen, mesodermalen oder entodermalen Differenzierungstendenzen. Die heutigen Kenntnisse über die charakteristische Musterbildung im axialen Teil des Mesoderms und in der überliegenden Neuralplatte scheinen in guter Übereinstimmung zu sein mit den des morphogenetischen Feldbegriffs. Dennoch wird benachdruckt, dass, falls das gegenwärtige morphogenetische Feldbegriff nicht weiter biochemisch und biophysisch gekennzeichnet werden kann, anstatt dessen ein anderer Begriff entwickelt werden muß, der schärfer definiert werden kann und besser zugänglich ist für experimentelle Analyse. Sich absolut setzende Wahrheiten blenden die Wahrnehmung aus, dass wir in einer falschen Welt mit falschem Bewusstsein leben. Der Poet Schiller mokiert sich in seinem Gedicht "Die Taten der Philosophen" (1795) hierzu. In der Schlusszeile spricht er die Krafte an, die aus dem Leben alle Wesen und in besonderer Weise den Menschen bestimmen: HUNGER und LIEBE. Vorliegende (...) Arbeit ist der Versuch einer Theorie des poetischen Menschen aus diesen Urkraften des Lebens. Es ist der Versuch der Wiedervereinigung von KUNST und Mensch und Absage an alle Wahrheits-Ideologien. Der Begriff “Wille zum Sinn” ist bei Frankl ein Schlüsselbegriff. Frankl überschritt die Grenzen der Psychologie, um die menschliche Existenz besser zu verstehen, die Einzigartigkeit der Person des Menschen, seine Freiheit und, wie er es selber benennt, sein “noogenes Wesensniveau”. Er stellt sich die existenzielle Frage, die der Mensch sich selber stellt. Um den Begriff “Wille zum Sinn” besser zu verstehen, brauchen wir eine breitere Erklärung. Da ist der Wille, das Streben, die Bemühung des Menschen um eine sinnvolle Existenz. Der (...) Sinn betrifft die Objektivität, die entdeckt werden soll. Weder gibt der Mensch sich den Sinn noch bildet er ihn sich selber. Der Mensch ist auf der Suche nach dem Sinn. Den Sinn entdeckt der Mensch mit Hilfe des Gewissens. Deshalb ist es nötig, ein Bemühen um die Kultivierung des Gewissens anzustreben. Die Frage nach dem Sinn stellt nicht der Mensch, sondern das Leben selbst. Das Leben fragt den Menschen in der Form der Wirklichkeiten. Eine Antwort des Menschen ist ein Übernehmen von Verantwortlichkeit. Der Sinn ist einzigartig und unwiederholbar. Das rührt her von der Einzig_artigkeit der Person und der Einmaligkeit der Situation. Der Sinn des Lebens ist in Bezug auf das Ziel. Der Mensch glaubt an den Sinn, ob er sich dessen bewu t ist oder nicht. Alles im Leben des Menschen hat einen grö eren Sinn. Das Leben kann sinnvoll durch die Arbeit werden, wenn diese Arbeit nicht egozentrisch ausgeführt wird. Einen Sinn des Lebens kann dem menschlichen Leben auch ein Erlebnis vermitteln oder gar ein Erlebnis der Liebe, welches das wertvollste Erlebnis dar_stellt. Dem menschlichen Leben kann man einen Sinn nicht nur durch Arbeiten und Erleben geben, sondern auch durch eine Fähigkeit zum Leiden, wenn der Mensch eine gute und menschenwürdige Position in der Situation des Leidens und des Schmerzes einnimmt. Mi erfolg des Lebens, Mangel an Lust oder an positiven Erlebnissen sind kein Zeichen für Sinnlosigkeit. (shrink)

Zur Zeit des Hl. Thomas von Aquin war es nicht leicht andere Religionen zu verstehen. Besonders auch deswegen nicht, weil die Verbreitung der Religion mit Macht und Krieg verbunden war. Aber gerade Thomas hat die sogenannte „Heidnische Lehre“ des Aristoteles in das Christentum eingeführt. In den Augen vieler orthodoxen Christen war dies ein unverzeihlicher Fehler. Mit dieser Lehre ist auch das Naturrecht (moralisch natürliche Gesetz) in die katholische Theologie eingeführt worden. Aber die Aristotelische Wiedereinführung (wenn nicht Revolution) in die christliche (...) Philosophie ist bis heute nicht ohne Hindernisse geschehen. Chesterton meint, dass Thomas korrigiert Platon mit Aristoteles, der die Dinge so genommen hat, wie er sie vorgefunden hat. Thomas nimmt die Dinge so, wie sie Gott geschaff en hat. Trotz allem ist es sehr wertvoll zu denken, dass die Wahrheit und der Glaube nicht im Widerspruch stehen können, und alles was die Wissenschaft ans Licht der Welt bringt, kann nicht im Widerspruch gegenüber dem Glauben sein. In den Werken des Hl. Thomas ist immer die Welt der positiven Schöpfung gegenwärtig. In dieser Arbeit möchten wir die Bedingungen der Zeit des Dialogs mit dem Islam und dem Judentum als Hintergrund für die Zusammenfassung der Summa contra Gentiles analysieren nach den Gedanken von M.-D. Chenu, O. H. Pesch, J. Weisheipl, K. G. Chesterton und anderen, die uns ein Bild über die nicht einfache aber trotzdem erreichbare Begegnung mit anderen Religionen zeigen und auch die Gefahr aufzeigen, die sich in einer nicht konsequenten Philosophie birgt. Die Chronologie umkehrend, bejaht Nietzsche nicht nur das, was überwunden werden muss, sondern zeigt auch, wie man der Vergangenheit Herr werden kann, indem man aus dem "Es war" der Geschichte ein "So wollte ich es" macht.
